Yes. Two-handed and thumbless releases load the core differently from a conventional grip, so the layout math changes. Our drilling layouts page explains Storm's Two-Handed Layout System, where the PAP is measured from a different reference and the layout is drawn in arcs, with a reference layout of 5 x 4 x 2.5.
